Protein G is a surface protein found in Group C and Group G Streptococcus bacteria. Although similar to Protein A, it exhibits distinct binding specificities. It exists in two forms: a 65 kDa variant (G148) and a 58 kDa variant (C40). This protein binds to both the Fab and Fc regions of immunoglobulins. Naturally, it also interacts with serum albumin, but since this is a common contaminant in antibody preparations, recombinant versions of Protein G have been engineered to eliminate the albumin-binding sites.

Protein G is widely used in antibody purification processes. The PurKineâ„¢ Protein G purification system is especially effective for isolating IgG from mammalian serum or other liquid sources. It excels at capturing monoclonal and polyclonal IgGs that may not bind efficiently to Protein A. For many mammalian IgG subclasses, such as human IgG3, mouse IgG1, and rat IgG2a, Protein G demonstrates stronger affinity than Protein A. Unlike Protein A, it does not bind to human IgM, IgD, or IgA.
