I often receive questions from students who ask us to order kits, saying that their samples were not stored properly. So what should they do? Improper storage of specimens in a refrigerator can cause issues, especially with long-term samples. For example, IgG in serum may polymerize into multimers, while AFP might form dimers. These changes can lead to high background signals or even false positives in indirect ELISA. If the sample is left for more than a day, the antigen or antibody activity might decrease, potentially resulting in false negatives. To avoid these problems, it's best to use freshly collected serum samples for ELISA. If immediate testing isn't possible, store the samples at 4°C for up to 5 days. For longer storage, cryopreservation at -20°C or -80°C is recommended. However, when thawing frozen samples, keep in mind that proteins may become concentrated or unevenly distributed.
